项目方向：CytosineDNAmethylationisoneofthemoststudiedepigeneticmarksineukaryotes,withestablishedrolesingenomeregulationandselfishDNAsilencing.Still,wedonotfullyunderstandhowmethylationinfluencestranscription.Inpart,thatlimitationislinkedtothewidespreadfocusonfewhighlycomplexmodelsystems,mostlymammalsorplants,withhighlycomplexmethylationpatterns.Inthisprojectwewanttofillthisgapbyunderstandingtheevolutionofcytosinemethylation“readers”,proteinscapableofexclusivelybindingtomethylatedcytosines.Tothisend,wewillusecomparativegenomicsandproteomicsonmultipleeukaryoteswithsimplermethylationlandscapes,occupyingkeypositionsinthetreeoflife.Theproteinsidentifiedinthepreviousapproachwillbethenvalidatedusingfunctionalgenomics(e.g.DAP-seq,ChIP-seq).ThisprojectwillrevealhowthevariousgenefamiliesthatareknowntohaverolesinDNAmethylationinterpretationevolved,andpotentiallywilldiscovernewproteinswiththiscapacity.Ultimately,understandinghoweukaryoticgenomesinterpretDNAmethylationisimportanttoclarifytherolesofthisepigeneticmarkingenomeregulation.Inturn,thesefindingswillhaveapplicationsinsyntheticbiologyapproaches.GiventhekeyrolesDNAmethylationhasinhumandisease,includingcancer,anevolutionary-informedmechanisticunderstandingofthisepigeneticmarkiscritical,asthevariousgenesandmechanismswelearnindivergenteukaryotescanbethenusedtoreadandinterpretourownepigenome.
大致要求:Applicationsareinvitedfromoutstandingcandidateswithorexpectingtoreceiveafirstorupper-secondclasshonoursdegreeandamastersdegreeinanarearelevanttotheproject(MolecularBiology,Bioinformatics,Biochemistry).Amastersdegreeisdesirable,butnotessential.ForthisPhD,anypreviousexperienceonmolecularbiologywork,fromcloningtoproteinpurificationwouldbehighlybenefitial.Furthermore,somebasicknowledgeofbioinformaticswouldalsobehelpful,althoughwearehappytoprovidetrainingonthisaspect.Theprojectwouldcombinewetlabandbioinformatics.
